Andrographis paniculata (Burm. f.) Nees is the whole grass or leaves of Andrographis paniculata (Burm. f.) Nees. 1. Pharmacological activity:
In a psycho-pharmacological study of Andrographis paniculata lactone. The extract showed significant alterations in behavioral patterns and decreased efficacy of spontaneous movements. The extract also prolonged the duration of pentobarbital-induced sleep and reduced body temperature in animal models of different experimental settings.
2. Anti-HIV virus:
A type I pharmacokinetic clinical trial including 13 HIV-positive patients and 5 HIV-uninfected healthy volunteers on andrographolide from Andrographis paniculata. The objectives were primarily to assess safety and tolerability, with secondary aims to assess its effect on plasma viral particle HIV-1 RNA levels and CD4(+) lymphocyte levels. None of the subjects used antiretroviral drugs in the trial. Those with liver and kidney malformations were excluded. The planned treatment regimen was a 3-week dose of 5 mg/kg body weight, rising to a 3-week dose of 10 mg/kg body weight, and finally a 3-week dose of 20 mg/kg body weight. A significant increase in mean CD4(+) lymphocyte levels (from 405 cells/mm(3) baseline to 501 cells/mm(3); p=0.002) was found in the HIV group following administration of 10 mg/kg of andrographolide. There was no significant quantitative change in mean plasma viral particle HIV-1 RNA levels during the experiment. Andrographolide may inhibit the impaired cellular circulation caused by the HIV virus, leading to increased levels of CD4(+) lymphocytes in HIV-1-infected patients.
